Data Documentation for Plama Quanterix HD-X/Simoa Neurology 4-plex E Dataset (Madison)

This record contains the README and data dictionary for the plasma Quanterix HD-X/Simoa Neuro 4-plex E data obtained locally.

Freeze

This documentation pertains to the May 2025 Freeze of the plasma Quanterix HD-X/Simoa Neurology 4-plex E- Madison dataset.

Origin

These multiplex plasma assay results come from the University of Wisconsin ADRC Biomarker Lab and were analyzed using the Simoa N4PE (GFAP, NfL, ABeta42, ABeta40) assay on the Quanterix HD-X platform.

Sample Selection

Samples are collected on an ongoing basis, starting August 3, 2011. Samples were stored in varying tube types depending on the study and date of collection.
For detailed information, refer to the WRAP and/or ADRC pre-analytic protocols for CSF and plasma collection, available on the Wisconsin Alzheimer's Program KnowledgeBase.

Sample and Participant Characteristics

This dataset includes 5483 observations on 1898 participants (480 ADRC; 1418 WRAP; 1280 female; 280 URG; 107 MCI and 21 Dementia at first visit; 105 MCI and 31 Dementia at last visit; 8 participant(s) are aged 90+, of those not: mean first and last ages of 62.2 and 66.9, respectively; mean first and last days since baseline of 2257 and 3996, respectively).

Per data sharing agreements with the Oneida nation, data from Native American participants are not included in this dataset. 

Analytes

  • Glial fibrillary acidic protein (GFAP)
  • Neurofilament light (NfL)
  • Amyloid beta 42 (AB42)
  • Amyloid beta 40 (AB40)

Dynamic Range

  • GFAP: 0 - 20,000 pg/mL (with 4x dilution)
  • NfL: 0 - 2,000 pg/mL (with 4x dilution)
  • AB42: 0 - 100 pg/mL (with 4x dilution)
  • AB40: 0 - 280 pg/ml (with 4x dilution)

All the concentrations reported for these analytes are in pg/mL.

Methods

Sample analysis was performed between March 25, 2024 and June 25, 2024 using a single kit lot (#503864).
