Data Documentation for Quanterix/ALZpath pTau217 Dataset (Madison)
Freeze:
This documentation pertains to Freeze 2024_November of the Quanterix/ALZpath - Madison dataset.
Origin:
These plasma assay results come from the University of Wisconsin ADRC Biofluid Lab and were analyzed using the ALZpath pTau217 v2 assay on the Quanterix HD-X.
Sample Selection:
Samples were collected between Aug 2011 and July 2023. Samples were stored in varying tube types depending on the study and date of collection.
This dataset includes 5267 observations on 1873 participants (480 ADRC; 1392 WRAP; 1269 female; 258 URG; 84 MCI and 15 Dementia at first visit; 131 MCI and 35 Dementia at last visit; 8 participant(s) are aged 90+, of those not: mean first and last ages of 62.7 and 67.2, respectively; mean first and last days since baseline of 2286 and 3934, respectively).
Per data sharing agreements with the Oneida nation, data from Native American participants are not included in this dataset.
Across all cohorts, two kit lots were used. 5,082 samples were analyzed using kit lot# 999008. 309 samples were analyzed using kit lot# 999024. A bridging study was performed using N = 35 samples to determine the relationship between lots. The Pearson correlation coefficient was 0.97, p < 2.2e-16.
Linear regression was used to standardize the data from lot# 999024 to lot# 999008 (y = 0.65514x + 0.02667).
Analytes:
- Phosphorylated tau 217
Dynamic Range:
- 0.00732 - 30.0 pg/mL (with 3x dilution)
Funding/Acknowledgments:
Papers including data obtained from these assays should acknowledge NIH R01 AG027161 (the Wisconsin Registry for Alzheimer's Prevention) and/or NIH P30 AG062715 (the Wisconsin Alzheimer's Disease Research Center).
A manuscript is under development under the guidance of Sterling Johnson and Henrik Zetterberg regarding the validation of plasma pTau217 to amyloid and tau PET, longitudinal characterization, and relationship with cognition.
